Loratadine #CAS79794-75-5
It is a stable chemical compound of a tricyclic aromatic non-sedating antihistamine. At room temperature, this substance appears as a white or off-white crystalline powder. Its structure contains a tricyclic core of a chlorinated benzodipyridine ring, and is coupled with a piperidine ring containing a neutral amine carbamate group. This specific low polarity and weak basic structure makes it extremely difficult to penetrate the blood-brain barrier. In terms of solubility chemistry, due to the presence of a large number of hydrophobic aromatic rings, it is almost insoluble in water, but readily soluble in organic solvents such as methanol, ethanol, acetone, and chloroform. Under normal conditions, this substance has good thermal stability and light protection stability, but it is strictly prohibited from contact with strong oxidants. In the body or in strong acid-base environments, the amine carbamate bond within the molecule is highly prone to hydrolysis and decarboxylation reactions, removing the carboxyethyl group and efficiently converting into a more active and slower-metabolizing derivative, desloratadine.
Application Of Loratadine
antinflammatory, analgesic, antipyretic.Histamine H1 receptor antagonist; antihistamine.A nonsedating-type histamine H1-receptor.Loratadine, is used as a peripheral histamine H1 receptor agonist. It is also used as an orally active antiallergic agent. It is an important raw material and intermediate used in Organic Synthesis, Pharmaceuticals, Agrochemicals and Dyestuff.
The highly economically valuable bulk generic drug raw materials (API) and the core products of pharmaceutical fine chemicals. In modern pharmaceutical industry, it is widely used on a large scale for the production of tablets, capsules, oral solutions, as well as rapid disintegration tablets and chewable tablets for children and patients with swallowing difficulties. It is one of the most crucial pillars in the global supply chain of anti-allergic and antihistamine drugs. During the fine synthesis stage of raw materials, its industrial production line highly relies on large-scale crystal control crystallization processes (such as controlling the solvent ratio and cooling crystallization rate) to ensure that the powder produced industrially in large quantities has the dissolution rate, tablet hardness and fluidity that meet the strict pharmacopoeia standards. Moreover, due to its high lipid solubility and excellent formulation compatibility, it is widely used in the industrial applications of modern new drug delivery systems (DDS), such as pharmaceutical microencapsulation, hot melt extrusion (HME), and nano-crystal solubilization, to develop new generation of composite anti-allergic preparations with longer duration, sustained release and higher bioavailability.
